No Amtrak to Big Bend or Guadalupe.
You can get to Alpine on Amtrak and then you will need to rent a car there and then it is a two hour drive south. Guadalupes would mean probably renting a car in El Paso then an equally long drive...

Not sure if it meets your thru hiker requirement. I know of at least one report of a Kalais being used for a PCT thru.
